Histochemical Localization of Carbohydrates in Morphological Stages of Developing Human Minor Salivary Glands: A Comparative Study with Cytoskeletal Markers International Journal of Morphology
Rêgo,Moacyr J. B. M; Cavalacanti,Carmelita L. B; Beltrão,Eduardo I. C; Sobral,Ana Paula V.
Carbohydrates play a critical role in many cellular processes like disease, growth and development. In this work lectins, proteins that recognizes carbohydrate free or conjugated, were used as histochemical probes for carbohydrates localization in developing human minor salivary gland. Immunohistochemistry for traditional cytoskeleton markers (Cks 7, 8, 13, 14, 19, SMA and Vimentin) was performed and then compared whit lectin histochemistry for PNA, WGA, ConA and UEA-I, specifics for D-galactose, N-acetyl-glucosamine, glucose/mannose and L-fucose respectively. For this, specimens were obtained from tongues and lips of 15 human foetuses at 10-28 weeks of gestation. None of immune cytoskeleton markers were identified in the first stage of development...

Histomorphological and quantitative immunohistochemical changes in the rat pancreas during aging Biocell
Riccillo,F.L.; Bracamonte,M.I.; Cónsole,G.M.; Gómez Dumm,C.L.A..
Although the endocrine pancreas is the purpose of several deep investigations, morphological data referred to the effect of aging on the gland are not homogeneous. The purpose of the current work was to analyze the changes occurring in the pancreas of aged rats, with especial reference to the islet cell populations. Six young (Y), old (O) and senescent (S) male Sprague-Dawley rats were used. The pancreas tails were processed for light microscopy and studied by means of routine stains as well as by immunohistochemical identification of insulin-, glucagon-, somatostatin-, and pancreatic polypeptide- secreting cells (Dako Envision System, DAB as chromogen). A progressive pancreatic histoarchitecture distortion was found among the aged animals. Even when the...

Human testis-expressed sequence 101 is limitedly distributed in germinal epithelium of testis and disappears in seminoma Biol. Res.
Shen,Cong-Cong; Kang,Yu-Huan; Yu,Lin; Cui,Dan-Dan; He,Yi; Yang,Jin-Liang; Gou,Lan-Tu.
BACKGROUND: Testis-expressed sequence 101 (TEX101) was found to be highly expressed in testis and involved in acrosome reaction in previous studies. Recently, the metastasis suppressor function of TEX101 in cancer was disclosed, but the comprehensive investigation of its expression has rarely been reported. In this study, the expression features of TEX101 in normal human organs and seminoma were systematically analyzed. RESULTS: Immunohistochemistry demonstrated intense staining of TEX101 in human testis tissues; however, its expression in 27 other types of normal human organs, including the ovary, was negligible. Higher expression of TEX101 was observed in the spermatocytes and spermatids of the testis, but relatively lower staining was detected in...

Identification of the autophagy pathway in a mollusk bivalve, Crassostrea gigas ArchiMer
Picot, Sandy; Faury, Nicole; Arzul, Isabelle; Chollet, Bruno; Renault, Tristan; Morga, Benjamin.
The Pacific oyster, Crassostrea gigas, is a mollusk bivalve commercially important as a food source. Pacific oysters are subjected to stress and diseases during culture. The autophagy pathway is involved in numerous cellular processes, including responses to starvation, cell death, and microorganism elimination. Autophagy also exists in C. gigas, and plays a role in the immune response against infections. Although this process is well-documented and conserved in most animals, it is still poorly understood in mollusks. To date, no study has provided a complete overview of the molecular mechanism of autophagy in mollusk bivalves. In this study, human and yeast ATG protein sequences and public databases (Uniprot and NCBI) were used to identify protein members...

Paiotti,A.P.R.; Artigiani Neto,R.; Forones,N.M.; Oshima,C.T.F.; Miszputen,S.J.; Franco,M..
Ulcerative colitis (UC) is a disease of the colon and rectum characterized by a nonspecific chronic inflammation mediated by the concerted response of cellular and humoral events. Prostaglandins are synthesized by cyclooxygenase (COX)-1 and -2 and exhibit both pro- and anti-inflammatory activity. To evaluate COX-1 and COX-2 immunoexpression in 42 cases of UC and to correlate it with clinicopathological parameters, COX-1 and COX-2 expression was investigated by the immunohistochemistry method. Only patients with all pertinent clinical and evolutive data as well as with adequate biopsy material were included in the study. Fifteen samples of colorectal adenocarcinoma and 14 of large bowel with no histological changes were used for positive and negative...

Immunohistochemical Detection of BMP-2 and BMP-4 in the Developing Neural Tube and Spinal Cord of Human Embryos International Journal of Morphology
Namm,Aimar; Arend,Andres; Aunapuu,Marina.
The role of bone morphogenetic proteins (BMP-s) in the development of the nervous system has been widely studied on avian and rodent embryos. Human embryos have rarely been available for detection of BMP expression. In this study 39 human embryos of Carnegie stages (CS) 10-20 were investigated. The embryos were fixed in paraformaldehyde, embedded in paraffin and sectioned serially in transverse direction. BMP-2 and BMP-4 protein expression in the developing neural tube and the caudal spinal cord was determined by immunohistochemistry. Our data show that BMP-s tend to be more expressed in the neural tube in earlier stages; in particular, BMP-4 staining was found to be higher at CS10 compared to CS20. More detailed analysis was performed on embryos of...

Immunohistochemical detection of virus through its nuclear cytopathic effect in idiopathic interstitial pneumonia other than acute exacerbation BJMBR
Santos,G.C. dos; Parra,E.R.; Stegun,F.W.; Cirqueira,C.S.; Capelozzi,V.L..
Idiopathic interstitial pneumonias include complex diseases that have a strong interaction between genetic makeup and environmental factors. However, in many cases, no infectious agent can be demonstrated, and these clinical diseases rapidly progress to death. Theoretically, idiopathic interstitial pneumonias could be caused by the Epstein-Barr virus, cytomegalovirus, adenovirus, hepatitis C virus, respiratory syncytial virus, and herpesvirus, which may be present in such small amounts or such configuration that routine histopathological analysis or viral culture techniques cannot detect them. To test the hypothesis that immunohistochemistry provides more accurate results than the mere histological demonstration of viral inclusions, this method was applied...

Immunohistochemical expression of B and T-lymphocytes and TGF-β in experimentally transplanted canine venereal tumor Ciência Rural
Silveira,Ana Carolina Trompieri; Gerardi,Daniel; Mouro,Juliana Vitti; Costa,Mirela Tinucci; Alessi,Antonio Carlos.
The tumor/host relationship may have a determining role in the progression or remission of a tumor. Greater infiltration of leukocytes into tumors has been associated with a better prognosis, although controversy regarding whether these cells have a central role in antitumor immunity still exists. Canine transmissible venereal tumor (TVT) is an experimentally transplantable type of tumor that has been used as an experimental model for the tumor/host relationship. The aim of this study was to evaluate the infiltration of T-lymphocytes (CD3, CD4, CD8) and B lymphocytes (CD79-α) and the expression of the cytokine TGF-β in TVT, by means of immunohistochemistry (ABC method). The experimental tumors were composed of puppies that developed TVT after...

Immunohistochemical Overexpression of MAP-2 in the Cerebral Cortex of Rabies-Infected Mice International Journal of Morphology
Hurtado,Andrea P; Rengifo,Aura Caterine; Torres-Fernández,Orlando.
The microtubule-associated protein MAP-2 is an integral part of the cytoskeleton and plays an important role in neural morphogenesis. This protein is an essential component of the dendritic cytoskeleton, especially in the adult brain, and its expression can be altered under experimental or pathological conditions. The purpose of this study was to evaluate the effect of infection with the rabies virus on MAP-2 immunoreactivity in the cerebral cortex of mice. The mice were inoculated with the rabies virus and the animals were sacrificed when the disease reached its advanced stage, together with uninfected animals of the same age. The brains were extracted after being previously perfusion-fixed with paraformaldehyde; coronal sections were obtained with a...

Immunohistochemical Study of Aquaporin 1 and 2 in the Reproductive Tract of the Bactrian camel (Camelus bactrianus) International Journal of Morphology
An,Fengli; Wang,Jianlin.
Spermatogenesis is associated with considerable fluid secretion or absorption in the male reproductive tract. Aquaporins (AQPs) are membrane protein channels that allow the rapid movement of water through epithelium. In the present study, immunohistochemistry was utilized to localize the expression of AQP 1, AQP2 in the testis and prostate of adult bactrian camel (Camelus bactrianus). Results show that AQP1 have intense reaction in rete testis, efferent ducts, vessels, seminiferous duct and in the prostate, AQP2 was found minor expression in the rete testis, vessels and prostate, which suggesting that AQP1 may have the main role in the absorption of the large amount of testicular fluid in male camel reproductive tract. Investigations of AQPs biology in...
